Effect of Hydrocortisone on the Utilization of Tritiated Thymidine for Skeletal Growth in the Rat.

چکیده
Thymidine is a specific precursor of deoxyribonucleic acid (DNA) and is utilized for chromosomal replication by the nuclei of cells when they double their DNA content in interphase before cell division (Reichard and Estborn, 1951; Friedkin, Tilson, and Roberts, 1956; Gall and Johnson, 1960; Mirsky and Osawa, 1961). Autoradiography after the administration of thymidine labelled with tritium (3H-thymidine) to experimental animals is now established as a valuable tracer of DNA synthesis in actively dividing cells, for the energy of the beta radiation emitted by tritium (0 018 MeV) is sufficiently low to allow precise localization of the label in single nuclei that are preparing to divide. Furthermore, the time taken for DNA synthesis is considerably longer than that required for actual nuclear division, so that a 3H-thymidine labelling index becomes a more sensitive indicator of cell renewal and proliferation than conventional assessments of the mitotic index (Cronkite, Bond, Fliedner, and Rubini, 1959; Leblond, Messier, and Kopriwa, 1959; Crane and Dutta, 1963). In the experiments described in this paper, we have used this technique to identify the various cells in normal rat skeletal tissues that utilize labelled thymidine and have measured the depression in the labelling index that results from treatment with hydrocortisone.

متن کاملA survey on the influence of intra- peritoneal injection of hydrocortisone in the rate of intra abdominal adhesion bands in rats
Occurrence of adhesions after pelvic and abdominal surgery is a well known problem. Previous surgical procedures are by far the most common cause of intra abdominal a adhesion. In over 10℅ of infertile women, adhesion formation is the main cause of their problem. Moreover postoperative adhesions are the most common cause of intestinal obstruction in the Western world. Although the cause of ...
متن کاملRole of mineralocorticoid receptors in the mediation of acute effects of hydrocortisone in isolated ischemic rat heart
Introduction: Cardiac preconditioning is an important method to reduce the damage caused by prolonged ischemia. Previous studies have shown that corticosteroids have protective effects on the heart, however at high concentrations this effect may be reduced with unknown mechanisms. We hypothesize that the contradictory effects of hydrocortisone at high concentration may be mediated via minera...
متن کاملEffect of acute tritiated water (HTO) exposure on Maze learning in adult Swiss albino mice
Background: Brain is the most important organ as it controls and co- ordinates all the activities of our body. Reports on neuroethological investigations in mice particularly associated with learning and memory after radiation exposure are very scanty. Hence, present investigation is an attempt to examine the effects of low dose tritiated water (HTO) acute exposure on adult Swiss albino mice in...
متن کاملThe effect of bone marrow mesenchymal stem cells on recovery of skeletal muscle after neurotization surgery in rat
Objective(s): When the nerve is injured near its entrance to the muscle belly, we cannot perform conventional methods. One useful method in such a situation is neurotization surgery. In this study, Bone marrow mesenchymal stem cells (BMSCs) implanted into the paralyzed muscle after neurotization surgery. These cells can stimulate axon growth and motor endplate formation, also prevent muscle atr...
